Elektrochemische Kombinatorik als Grundlage für die Entwicklung biologischer Assays

Die vorgelegte Arbeit veranschaulicht die Entwicklung eines elektrochemischen Robotiksystems für die Untersuchung biologischer Fragestellungen. Elektrochemische Experimente können vollautomatisch im Mikrotiterplattenformat durchführt werden und sind kompatibel zu anderen Hochdurchsatztechnologien.

Die Breite der möglichen Assays wurde am Beispiel der Herstellung und Charakterisierung von Biosensoren im 24- und 96-Well-Format sowie an der Untersuchung von Redoxmediatoren im 384-Format dargestellt. Als zellbasierter Assay wurde erstmals die vollautomatische, elektrochemische Untersuchung der örtlichen und zeitlichen Aspekte der Ausschüttung von Stickstoffmonoxid (NO) aus biologischen Zellen in Echtzeit gezeigt. Es wurde die Auswirkung des vaskulären Endothelwachstumsfaktors (VEGF) auf die NO-Signaltransduktion in humanen spontan transformierten Endothelzellen der Nabelschnurvene (T-HUVEC) untersucht.
