Inhalt der Arbeit:
Aim of the project was the development and application of suitable technologies for the multiple and combinatorial solid phase synthesis of transfections lipids. The strategy was based on the utilisation of trityl and vinyl sulfone resins as the solid support. Lipid libraries were synthesized and their transfection properties were screened. This strategy allowed to generate a diversity of new types of lipids whose structures were broadly varied. Severals new classes of transfection lipids were discovered by high throuput screening. The best members are 15 times more efficient than the well known cationic lipid DOTAP while having comparable toxicities.
